Course-Based Undergraduate Research Experiences (CUREs) are often heavily constrained and lack the clinical relevance needed to inspire pre-health majors. To address these issues, we developed BUDDY (Bioinformatic Utility for Diagnostic Discovery in Yeast), an accessible bioinformatic web app. BUDDY transforms a precision medicine bottleneck, the millions of uncharacterized human Variants of Uncertain Significance (VUS), into an opportunity for student-led inquiry. While yeast is an excellent model for functionally testing conserved human variants, executing the bioinformatics needed to identify target variants and genes would consume limited teaching lab sessions. BUDDY eliminates this hurdle by seamlessly retrieving variants, identifying orthologs, aligning sequences, rendering 3D protein overlays, integrating machine-learning pathogenicity predictions, and automating CRISPR/Cas9 oligonucleotide design. Finally, BUDDY utilizes multimodal AI to help design cost-efficient, safety-conscious experiments, typically by leveraging established yeast phenotypes to test if a VUS causes conditional growth defects linked to the target gene.
