In this workflow students use the recombineering method of CRISPR gene editing to replace the LacZ coding sequence with a GFP coding sequence in E. coli cells. Students run through all the steps from purifying plasmid and linear DNA, electroporating E. coli bacteria and then scoring, and interpreting results. Then they modify the guide RNA plasmid to target a different region of the LacZ coding sequence and repeat the workflow, comparing results between different guide RNA plasmids. At Shoreline College this workflow is integrated into a yearlong Biotechnology lab technician training program. However, the whole workflow or sections of it could be integrated into a broader range of courses for different college programs. Your feedback will inform future development and dissemination approaches for the workflow.
